Changes of metabolic parameters according to aging in p47phox-deficient mice

Abstract
Background : The metabolic syndrome (MetS) is a constellation of factors apparently associated to an increased
cardiovascular risk, and with a reported increasing prevalence in parallel with aging process. Oxidative stress is one of
the cause linking aging and MetS. Here, we studied the role of p47phox, a key enzyme of NAD(P)H oxidase, in change
of metabolic parameters and renal oxidative stress according to aging.

Methods : Mice deficient in p47phox (on C57BL/6 background) of 5-month-old and 16-month-old were used. Heart and kidney wet weights were measured. Serum cholesterol, glucose, and high sensitivity C-reactive protein (CRP) activity were measured. The activity of NAD(P)H oxidase was assessed by lucigenin-enhanced chemiluminescent detection.

Results : C57BL/6 wild-type (WT) developed higher body weight compared to p47phox-/- mice (43¡¾4.8 vs. 38¡¾6.5gm) as measured at 16-month-old. Cardiac and renal hypertrophy corrected by body weight in both young and old groups showed no difference. Total serum cholesterol was increased in 16-month-old WT by aging, but was significantly reduced in 16-month-old p47phox-/- mice (p<0.01 vs. WT). No significant changes of serum glucose and CRP activity were seen in 5-month-old and 16-month-old WT and p47phox-/- mice. Aging did not show increase NAD(P)H oxidase in kidney of WT and p47phox-/- mice.

Conclusions : Deficiency of the NAD(P)H oxidase subunit p47phox contributes to less increase of body weight and to decrease total serum cholesterol in a ROS-independent manner during the aging process.
